G protein-coupled estrogen receptor in GtoPdb v.2023.1

چکیده

The G protein-coupled estrogen receptor (GPER, nomenclature as agreed by the NC-IUPHAR Subcommittee on [26]) was identified following observations of estrogen-evoked cyclic AMP signalling in breast cancer cells [2], which mirrored differential expression an orphan 7-transmembrane GPR30 [6]. There are both cell-surface and intracellular GPER [29, 34]. Selective agonist/ antagonists for have been characterized [26]. Antagonists nuclear receptor, such fulvestrant [11], tamoxifen 34] raloxifene [25], well flavonoid 'phytoestrogens' genistein quercetin [18], agonists GPER. Reviews pharmacology published roles (patho)physiological systems throughout body (cardiovascular, metabolic, endocrine, immune, reproductive) also reviewed [26, 27, 20, 17, 9]. GPER-selective agonist G-1 is currently Phase I/II clinical trials (NCT04130516).

G-protein Coupled Receptor Dimerization

A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...

G protein-coupled receptor 30: estrogen receptor or collaborator?

More than 800 G protein-coupled receptors (GPRs) are transcribed in the human genome (1). For many of these receptors, no endogenous ligand has been found, and, therefore, they are currently designated as orphan receptors. One such protein, GPR30, has been proposed to be an estrogen receptor (ER) (2, 3). GPR30 has been localized either to the cell plasma membrane (2) or to the endoplasmic retic...

G protein-coupled estrogen receptor regulates mammary tumorigenesis and metastasis.

UNLABELLED The role of 17β-estradiol (E2) in breast cancer development and tumor growth has traditionally been attributed exclusively to the activation of estrogen receptor-α (ERα). Although targeted inhibition of ERα is a successful approach for patients with ERα(+) breast cancer, many patients fail to respond or become resistant to anti-estrogen therapy. The discovery of the G protein-coupled...

GPR30: A G protein-coupled receptor for estrogen.

Estrogen is a critical steroid in human physiology exerting its effect both at the transcriptional level as well as at the level of rapid intracellular signaling through second messengers. Many of estrogen's transcriptional effects have long been known to be mediated through classical nuclear steroid receptors but recent studies also demonstrate the existence of a 7-transmembrane G protein-coup...

G-protein Coupled Estrogen Receptor (GPER/GPR30) and Women's Health

Estrogen - the female sexual hormone playing the most important role - plays a physiologically significant role, not only regulating in cell signals with second messenger but also being active in regulating transcription. Estrogen receptor (ER) which is a protein accepting estrogen not only play the role of a transcription factor combining with other genes to regulate their activity like other ...
